G protein coupled receptor kinases (GRKs) play an important role in homologous desensitization of GPCRs. Eugenia V. Gurevich pointed out: GRKs appeared in the early evolution of eukaryotes. At that time, a kinase structure similar to the ribosomal protein S6 kinase was inserted into a ring structure of the RGS domain, resulting in the formation of GRKs. GRKs form two branches in multicellular animals, one branch includes GRK2 and GRK3, and the other includes all remaining GRKs. Among vertebrates, GRKs are mainly divided into two categories: GRK1-GRK7 and GRK4-GRK5-GRK6. One of the two ancient branches was discovered as early as the period of the silkworm. Several protists, two oomycetes and unicellular brown algae contain a GRK-like protein, which indicates that the kinase structure has been inserted into the RGS domain long before the multicellular organisms originated. The two GRK families obtain different structural units at the N-terminus and C-terminus, and these structural units are mainly responsible for cell membrane recruitment and receptor coupling.

Therefore, the results of the study indicate that GRKs appear significantly earlier than animals and spread rapidly in post-animals. Of course, this is probably due to the rapid regulation of signals by fast-moving animals.
