Ultrasonic Cell Disruption

Osmotic Shock Crushing
Osmotic shock is a gentler method compared to others. It works by placing cells in a hypertonic solution, such as a concentrated glycerol or sucrose solution, causing water to move out of the cells and leading to shrinkage. Once equilibrium is reached, the solution is rapidly diluted or the cells are transferred to a hypotonic environment. This sudden change in osmotic pressure causes water to rush back into the cells, making them swell and eventually burst. This technique is ideal for cells with more flexible membranes.
Chemical Disruption
Chemical methods involve using reagents like acids, bases, detergents, or organic solvents to dissolve the cell membrane or extract intracellular materials. These agents can weaken or destroy the structural integrity of the cell, allowing for the release of cellular contents. This approach is often used when other mechanical methods may damage sensitive components. However, care must be taken to avoid over-treatment, which could denature proteins or degrade nucleic acids.
Enzymatic Lysis
Enzymatic lysis uses specific enzymes to break down the cell wall, making it easier to access the cell's interior. For example, lysozyme is effective against Gram-positive bacteria, while Gram-negative bacteria require additional agents like EDTA to enhance the effect. In eukaryotic cells, different enzymes such as cellulase or protease may be used depending on the cell type. This method is highly selective and useful when preserving the activity of intracellular products is important.
Freeze-Thaw Method
The freeze-thaw method involves repeatedly freezing and thawing cells to induce rupture. When cells are frozen, ice crystals form inside, causing the cells to expand and break apart. Thawing further weakens the cell membrane, increasing permeability. This technique is especially suitable for cells with fragile walls and is often used in small-scale experiments where mechanical disruption might be too harsh.
